I don't know anything about Essen but if you are going to Tübingen and you like chocolate - Ritter sport headquarters and factory are located about 30 minutes by car south of Stuttgart in the village of Waldenbuch on the way to Tübingen. There is also a modern art museum next to Ritter sport

If you are driving from Holland Rhine region would be good, Bacharach and St Goar would be good choices. St Goar has a castle but Bacharach is quainter. Both have areas to wander or hike.

Also across river Marksburg castle.

Right now Stuttgart Stuttgarter Frühlingsfest going till may 9. I would stay Esslingen am Neckar pretty town.
Can take S1 bahn to Stuttgart center Modern art - Kunst or Bad Cannstatt - festival and
Esslingen am Neckar is close to freeway & easy to get to
Tübingen and Bebenhausen Abbey . Esslingen am Neckar freeway closer to next stop Fuessen or Garmisch than Tubingen.

On the way to Fuessen or Garmisch I would make a stop at Ottobeuren
abbey.

It is hard to decide between Hohenschwangau(where castles are) or Garmisch-Partenkirchen. Things to visit 2 castles Hohenschwangau or you can just take bus to bridge looks over valley (depends on weather) Linderhof castle. I prefer because of both interior and grounds and outer bldgs and grotto. Then there is Ettal Abbey and Wiesrkirche and Rottenbuch. If you want to go to Hohenschwangau castles go in the morning. I would go to google maps get directions and figure your own map.

Rothenburg ob der tauber stay overnight 1 night to go on night watchman tour and go on ramparts next morning
And if you can squeeze Wurzburg 1 night.
You did not mention how much time you have so I am assuming 7 or 8 days. It would be good uf you had a little more time. You can also take route in reverse if coming from Denmark

It's not clear if you only have a week, but if so, going to southern Germany and returining will take two days of your time. My wife use to drive from Luebeck to southern Baden-Wuerttemberg weekly- a long for the most part boring drive although there are plenty of interesting places off from the autobahn. Then visiting Holland also cuts into your time. Living in and loving Stuttgart, I like sandy12's ideas, but it's a long ways. Germany is full of interesting things to see and do, having over 30,000 castles and palaces alone. There's plenty to see in the north of the country which isn't that far from Denmark. www.schenckguide.com lists castles, palaces, gardens, historic houses and heritage sites for much of Europe, although most is on Germany where the sights are listed by German states. A long way from being even remotely complete, but a good start.
If you do decide to visit some of sandy12's suggestions, some useful websites for tourism for Baden-Wuerttemberg are : www.baden-wuerttemberg.de ; www.tourismus-bw.de ; www.schloesser-und-gaerten.de for the excellent castles, palaces, monasteries and gardens operated by this German state. A few other websites: our favorite afrea of Germany, the Swabian Alb ( www.schwaebischealb.de ); Lake Constance ( www.bodensee-tourismus.com ); Black Forest ( www.schwarzwald-tourismus.info ); Stuttgart ( www.stuttgart-tourist.de ); two lovely fairytale-like castles- www.burghohenzollern.com ; www.schloss-lichtenstein.de ; and a few lovely towns- www.ludwigsburg.de ; www.tuebingen.de ; www.esslingen.de ; www.meersburg.de ; www.badurach.de ; www.besigheim.de ; www.lauffen.de ; www.badwimpfen.de ; www.schwaebischhall.de ; www.vellberg.de ; www.ulm.de .
